Fourier ptychography for cell cultures imaging H/F

Sujet de stage

In microscopy, imaging large areas with high resolution has always been challenging. Either we look at small regions with fine details, or we look at large regions with coarse details.

Fourier ptychography (FP) is a computational method for synthesizing raw data into a high-resolution and wide field-of-view image through a combination of synthetic aperture and phase retrieval concepts.

Unlike conventional techniques, which trade resolution against imaging field of view, FP can achieve both simultaneously to produce high-throughput imaging of unstained transparent biological samples.

As an intern at CEA-Leti, you will have the opportunity to work in a world-renowned research environment. Our teams are made up of passionate and dedicated experts, providing a framework conducive to learning and collaboration. You will have access to cutting-edge equipment and top-notch research resources to carry out your missions.

 

 

During this internship, you will use an existing microscope setup with coded illumination patterns using LED array to acquire data from cell cultures samples.

 

You will implement the reconstruction FP algorithms using the available open-source code. You will work on further development of FP technique applications either by improving of the hardware setup or by adapting the reconstruction algorithms to cell-culture samples.

 

This technique can be eventually applied to the high-throughput screening of differentiating STEM cell culture samples in the scope of personalized cell therapy research.

Profil du candidat

This internship is intended for final-year engineering or Master 2 students with basics knowledge of Fourier optics and microscopy imaging techniques as well as moderate capabilities of coding in Python or equivalent programming language for image processing.

 

Good communication skills, curiosity and a taste for experimentation are appreciated.
